Tips for Agarose Gel Electrophoresis
The selection of buffers in agarose gel electrophoresis is contingent upon factors such as the size of the DNA fragment and the objectives of the post-migration analysis.
For isolating large DNA fragments (equal to or greater than 15,000 bp), the use of TAE buffer is recommended. Alternatively, if DNA extraction is not a concern, or recovery rates post-migration are not critical, TBE buffer is suitable, especially for smaller DNA fragments (1,000 bp or less).
The accompanying diagram illustrates the outcomes of applying an EzDNA Ladder to a 1.5% agarose gel utilizing EzRunTAE or EzRunTBE buffers, subjected to electrophoresis at 150V for 20 minutes using a Submerge-Mini. Notably, TAE buffer is well-suited for polymer separation, while TBE buffer is more appropriate for low molecular weight separation.
The WSE-1710 Submerge-Mini is a compact and efficient electrophoresis system with dimensions of 190mm (W) x 130mm (D) x 60mm (H) and a weight of 0.45kg. The gel tray comes in two sizes: Small (S) with dimensions of 54mm (W) x 60mm (L) and 4 wells, and Large (L) with dimensions of 110mm (W) x 60mm (L) and 2 wells. The 3mm / 5.8mm double-edged comb features 1mm thickness and is compatible with multi-pipets. The comb configuration includes 9 wells x 2 and 5 wells x 2 for the Small tray, and 22 wells and 12 wells for the Large tray. The system accommodates a buffer solution of 200 to 230mL. It operates on AC 100-120V, 50/60Hz input, with DC 50V / 100V / 150V output at 40W. The timer function allows for a range of 1 to 99 minutes, with a display showing the remaining time in LED format (Hour, min). The system ensures safety with features such as a sample comb, power indicators, and error displays for cover open (E3) and insufficient buffer (E5).
How to use
Apply a voltage to the electrophoresis chamber.
Allow the DNA to migrate through the gel.
Stop electrophoresis and visualize the DNA on the gel using an appropriate staining method, such as ethidium bromide or SYBR Green.
Pour the gel into the gel tray and allow to polymerize.
Load the DNA samples into the gel wells.
Fill the electrophoresis chamber with TBE buffer.
Prepare the polyacrylamide gel by mixing the acrylamide, bisacrylamide, and Tris-borate-EDTA (TBE) buffer solutions.
