Tips for Agarose Gel Electrophoresis

The selection of buffers in agarose gel electrophoresis is contingent upon factors such as the size of the DNA fragment and the objectives of the post-migration analysis.

For isolating large DNA fragments (equal to or greater than 15,000 bp), the use of TAE buffer is recommended. Alternatively, if DNA extraction is not a concern, or recovery rates post-migration are not critical, TBE buffer is suitable, especially for smaller DNA fragments (1,000 bp or less).

The accompanying diagram illustrates the outcomes of applying an EzDNA Ladder to a 1.5% agarose gel utilizing EzRunTAE or EzRunTBE buffers, subjected to electrophoresis at 150V for 20 minutes using a Submerge-Mini. Notably, TAE buffer is well-suited for polymer separation, while TBE buffer is more appropriate for low molecular weight separation.

How to use

5

Apply a voltage to the electrophoresis chamber.

6

Allow the DNA to migrate through the gel.

7

Stop electrophoresis and visualize the DNA on the gel using an appropriate staining method, such as ethidium bromide or SYBR Green.

2

Pour the gel into the gel tray and allow to polymerize.

3

Load the DNA samples into the gel wells.

4

Fill the electrophoresis chamber with TBE buffer.

1

Prepare the polyacrylamide gel by mixing the acrylamide, bisacrylamide, and Tris-borate-EDTA (TBE) buffer solutions.
