If the cells are not dislodging after 5 minutes, it may be due to one of the following reasons:

  • The cells are not properly attached to the dish.
  • The trypsin solution is too old or has been diluted too much.
  • The cells are resistant to trypsin digestion.

If you are having trouble dissociating the cells, you can try the following:

  • Increase the incubation time up to 10 minutes.
  • Use a fresh batch of trypsin solution.
  • Try using a different type of trypsin, such as TrypLE Express.
